Interface can have full loopback routing capability (RME) a fixed one (MOTU M4, Audient.) or none. Then you have your outputs playing as inputs and you can further mix them with something like microphone, which is an input anyway.

You have to route output to the input of your interface. software needs inputs and most of these SWs have one input only. You have to think of it this way - when you play audio (at once) from your software (DAW, browser, player, standalone instrument.) it goes to your interface outputs.
